Professor Joshua Lederberg
Department of Genetics
The University of Wisconsin
Madison 6, Wisconsin
I am surprised to notice that your letter arrived here nearly two months ago. I have not had any special reason for postponing my answer just the usual one of having been busy with various jobs.
I have often pointed out, as you mention, that a crystal of a molecular substance can be considered as showing complementariness - each molecule is complementary in structure to the aggregate of the surrounding molecules. In the process of crystallization the molecules select the structure (the way of packing) that gives the greatest complementariness, and hence the greatest stabilization through intermolecular attraction.
One might ask, as you do, whether two different kinds of molecules, A and B, might not be able to pack together, in a crystal, better than the individual molecules can pack by themselves in the crystals of the separate pure substances. The answer is that this might happen sometimes and not other times. For example, when a racemic mixture of two optical isomers is allowed to crystallize, one obtains racemic crystals roughly half the time, and right-handed and left-handed crystals the other half of the time. In the same way, when salts are crystallized together, such as potassium chloride and magnesium chloride, one may obtain crystals of a double salt, or one may obtain two kinds of crystals, of the individual salts.
I have not so far made any reference to mixed crystals (solid solutions). I think that one can understand why mixed crystals in general form only between substances with very nearly the same molecular size and shape. A crystal of substance A, for example, has, when one molecule A is removed from inside it, a cavity into which this molecule fits neatly; in general a molecule B would not fit into this cavity, and hence in general A and B would not form solid solutions.
Sincerely yours,
Linus Pauling:W

Dear Mamma,
Happy New Year. Things are moving slowly. Christmas was quiet. New Year's was
quiet. Linda will be back soon & then perhaps she and & I can have the pudding, which I have
not eaten yet. Rumor says she was in good form in Carradale and got along like a house afire
with the Mitchison's. Similarities in the Mitchison-Pauling house holds have made her homesick.
Do you want this amplifier? My work is not as happy as it might be. I am unsure what is
wrong. I a m still living in my little room in Newnham. It is sad, but I am little there so it does
not much matter.
Much love,
(XXXXOOOO)google! Peter

16 January 1956
Dr. Richard T. Arnold
Alfred P. Sloan Foundation, Inc.
30 Rockefeller Plaza
New York 20, N.Y.
Dear Dr. Arnold:
I am writing to nominate Professor John D. Roberts for a grant for support of research by the Sloan Foundation, in the field of exploratory work on organic syntheses and reaction mechanisms.
For some time I did not think of nominating Professor Roberts, because I had the feeling that you were interested in younger men. However, I realize that Professor Roberts is only 37 years old, and that I had thought of him as older because he has such a fine reputation as a scientist. I think that you know Professor Roberts well enough for me to make only a rather brief proposal.
Professor Roberts would like to have some uncommitted funds to permit him to investigate rather long-shot possibilities in the field of organic chemistry - researches requiring rather more skillful and adventurous personnel than even the best graduate students, who tend to conservatism because of thesis requirements. He would like to have funds enough for a post-doctoral fellow, over a two or three year period, who might work on meta-stable organic substances obtainable only by unusual reactions, and preservable in substantial quantities at liquid air or liquid helium temperatures. This work might be done in part in collaboration with Professor Pellam. The grant required for this purpose would be about $6,000 per year.
At the present time Professor Roberts is carrying on research in organic chemistry with the aid of general funds of the California Institute of Technology, and also of the following special funds:
1. Carbonium ion rearrangements (l954-1956), supported by the Petroleum Research Fund of the American Chemical Society, about $8,000 per year. Application has been made for a terminal award, for the year 1956-1957.
2. Small-ring compounds (1955-1957), supported by the National Science Foundation, about $6,000 per year.
3. Fixation of nitrogen (1954-), supported by the John Simon Guggenheim Memorial Foundation, one grant of $15,000, to be spread out as desired.
In addition, Professor Roberts has applied to the Office of Naval Research for support of research in the fields of photochemistry and nuclear magnetic resonance, $10,000 per year for one to three years.
It is my opinion that Professor Roberts is so outstanding in originality that it is well worth while for him to be given support that he may use in carrying out investigations along imaginative and perhaps unpromising lines. His Guggenheim Fellowship grant is of this sort, but it will be used up before long, and Professor Roberts has some other ideas that he wishes to try out.
I think that Professor Roberts is one of the most promising, able, and original organic chemists in the country, and that he could be counted on to do something worth while with a grant made to him by the Alfred P. Sloan Foundation.
Sincerely yours,
Linus Pauling:W

Dear Mom,
I arrived with no difficulty, a little late. We came a strange way, over the ocean for a while, then up the Willamette valley. Saw signs of flood damage from the air, just Ike.
Monte was quite pleased with the bracelets; she wears them all the time. One guess what she gave me, the unfortunate thing is that I am more or less committed to wear the damn things, and her Pop apparently makes pretty awful ties. Oh well, life. The rush of the end of the semester is catching up with me, as I slowly realize how much I haven't learned. I have just been going over the German vocabulary, quite a job.
Monte and I are getting rather close, not immoral, but you know what I mean. Probably too close. I am pretty fond of her, and from the way that she talks I judge that she is pretty fond of me. She has started talking about marriage, pretty generally, asks vague questions as to my desires for the religious education of my children, etc. Certainly as far as I am concerned at this stage nothing will come of it, so don't you start worrying. However, I am pretty fond of her.
Jerry brought my razor cord when he came, apparently he found it and thought that I might like to have it.
I trust that you had a good drive down from Berkeley, and had a good time in Berkeley. What furniture did you buy? Love to Daddy and Milton.
Love,
Crellin

18 January 1956
Dear Peter:
Mama and I have spent about ten days in Berkeley. I gave several lectures there, one of them a sort of seminar in biology - except that there were about 500 people present. This talk was on the place of hypothesis in scientific research. Actually, it was mainly a discussion of the stochastic method.
Crellin was home for Christmas. Milton, our nephew, has come back from Philadelphia. He is planning to enter Pasadena City College next month, and he is living with us at present.
Mama and I are going to go to Berkeley again in February, for about ten days. Also, we are going to drive up to Arcata, near Eureka, in northern California, where I am to give a talk on early science in America, at the Humboldt Junior College, in connection with their celebration of the 250th anniversary of the birth of Benjamin Franklin.
Professor Corey and I are enthusiastic about plans that we have made for attacking the problem of the structure of a crystalline globular protein. I think that it is going to be a hard job, but that it can be done.
While we were in Berkeley we visited the Stanleys. I had dinner with them twice, the first time before Mama had come up (she had stayed in Pasadena for three days, in order to be with Crellin, before he started back to Reed). Professor Stanley mentioned to me that you were trying to get a fellowship to come to his laboratory to work on electron microscopy. He said that he felt that the laboratory would give you a postdoctoral fellowship, in case that you did not obtain an outside one. I don't know whether he thought that I would tell you this or not.
Dr. Dintzis, in a letter to me, mentioned a densitometer permitting x-ray diffraction spots on photographs to be measured to within 2%. I am not sure what sort of an instrument he had in mind. We make use almost entirely of visual estimates of intensities. Can you tell me how you measure intensities on your protein photographs? Is there a densitometer on the market that can be used for this purpose?
We are looking forward to your return. Mama thinks that we might come to see you get your doctor's degree next June, or perhaps just come for a trip.
Love from
[Linus Pauling]

Dear Daddy,
I am sorry about the mistakes and vagueness in my recent correspondence.
I think that I may get out of my three projections of myoglobin by locating the atoms of mercury in a compound of myoglobin with para choloro mercury benzene sulfonic acid. I think the projection the difference Patterson of which I am now awaiting will confirm my interpretation of the first difference Patterson, and I think I will be able to refine the difference structure of HgIn [handwritten note: n=2,3], which makes very large differences in the diffraction pattern.
John Edsall was around yesterday and today. He seems well. He talked to Howard Dintzis, but I do not yet know what they concluded. I have seen Jack Dunitz, and his wife and they are well, but searching for a place to live in London, which is quite difficult.
I have not worried much about next year, and am somewhat upset that Robley Williams will be in Cambridge next year, I could stay here, I think and work in the Molteno with Kenneth Smith, Roy Markham and Robley Williams.
Have you heard anything about your rotating anode X-ray tube? I have written to Holmes, but he has not yet answered. Our present seals have now run for well over 1000 hours in each of two tubes, and I am beginning to think that they might be practical.
Linda is sill in Edinburgh, and I think reasonably happy. I hear she was in good form over Christmas, and fitted in well with the Mitchison’s. I think she likes them.
Francis Crick, Jim Watson, and Don Caspar are all working hard on viruses. Rosie Franklin has some results on the differences between TMV and the A protein without RNA, and the reaction of the A protein with mercury. The present idea is that the RNA is at 40 A radius, and consequently there must be some protein inside the RNA chains. They also think there are 14 RNA chains, tilted at 45 to the virus axis, and a fourfold screw up each chain.
I think that a good convenient proportional counter spectrometer may be the best method of gathering three dimensional protein crystal diffraction data. It would be nice if the gadget were automatic, but that may be too much to ask.
I have not done my calculations using the co-ordinates that we took, but think I might try to make a little program for relaxation.
Much love,
Peter

Dear Lloyd:
I was surely pleased to receive your letter of 1 January and to know that you approved of my performance on the TV show "Youth Wants To Know."
I found the experience of appearing on the show very interesting. I had not known what the questions were that the boys and girls who participated would ask me. The show was kinescoped three days before, and I was pleased to see that there had been no editing whatever - it appeared over the air exactly as recorded.
My wife joins me in sending best wishes to you for the New Year.
Sincerely yours,

Peterhouse
Dear Mamma,
Thank you for your letter. I had begun to worry that you were not speaking to me anymore. I shall so it to Linda, who is, I think, returning to Cambridge within the week.
The joker in the local store says there is a three month delay in the arrival or delivery of QUAD amplifiers, and that consequently it should be ordered soon. He also suggests a British made RCA amplifier as being better and with sooner delivery. I do not know whether it is better or not, it does not look as nice.
I have some plans for a super duper gramophone, which I wish to make when I return. The man I want to work with will be in Cambridge. Perhaps I shall stay here for another year. I do not know that Stanley would agree to that, and there is a shortage of electron microscopes here. Perhaps I shall have to come back.
I think the Gray arm is probably not as good as ours, though I am not sure.
Linda’s boy is quite virginal. The Ph.D. ceremony takes three minutes, and I must do some work first. It will be good to see you though. I would like to go on a trip to the south in the summer. Perhaps we could all go to Majorca and work on the Nature of The Chemical Bond for a while. I understand it is quite pleasant for authors there, and I know a chap who works there as a chef or a tourist boat driver or something. He is not very interesting, and neither is Robert Graves, but they would do for a while.
Much love
Peter

24 January, 1956
Dear Daddy,
Thank you for your letter of the 18th. I am glad you are interested in the attack on crystalline globular proteins, as I think that probably the problem can be solved with a lot of work.
We have a micro-densitometer that uses a split light beam and uses the error difference between one beam through the photograph and the other beam through an optical wedge to drive the wedge up and down. There is a pen attached to the wedge. There are two mechanically connected stages, one with the film and the other with a sheet of recording paper, and a screw to drive them. It is quite good, and we use it exclusively. It traverses a straight line, however, and is therefore of greatest use with precession camera photographs, which have an undistorted mapping of the reciprocal lattice. We shall attempt to get as much three dimensional data as possible with precession photographs, though I do not know that we shall be entirely successful. It is not accurate to 2%, the film itself varies more than that, and we measure only peak height and make no attempt to integrate, and while this is reasonable over similar crystals similarly oriented, it will not be as satisfactory on upper layers. The machine was designed by Peter Walker at King’s College, London, and is manufactured more or less by Joyce, Loeble and Company in Newcastle. It costs about 2800 dollars, and needs being worked over after arrival. [Handwritten note: "If you are interested, perhaps we should enquire for you.” and “J L and Co. Ltd. Uline Lane Newcastle-upon-Tyne 1”]
I am of the impression that it is simpler to measure three dimensional spectra with a proportional counter, using converging beam, stationary crystal methods.
I am glad that you have talked to Stanley. I am a little worried about the fellowship, and a little worried that Robley Williams will be here. I must write him.
I should be glad to see you in the summer. If something does not appear in my difference Pattersons, I shall be a little stuck. I was very hopeful from one projection and the one dimensional Pattersons, but the second projection is not pretty. Howard Dintzis has a new heavy metal, however, and is making me some crystals. I am working on the orthorhombic form of Sperm Whale myoglobin, which I think is more suitable for three dimensional work than the monoclinic form.
While I think you would do well to have some densitometers around, the more I think about trying to get three-dimensional data on film, the more I feel a counter spectrometer is better. I understand that a two circle gadget to fit on the GE Powder spectrometer to the design of the Harker one is being made by Supper or someone. Holmes would be against buying such a thing and I think I would agree with him. However, perhaps his great big spectrometer would be suitable. The King’s people (Peter Walker) are working on flying spot integrating photometer, which will probably work and would be excellent for Weissenberg photographs and other distorted spot shape photographs.
Much love
Peter

27 January 1956
Dear Peter:
I am glad to learn some details about your work on myoglobin.
I am sure that something worth while can be learned from the three projections. Also, I feel sure that the determination of the structure of a crystalline protein will require that the phases of general reflections be determined, permitting a three-dimensional Fourier to be constructed.
I think that I told you that we offered Howard Dintzis a job. I do not know yet whether or not he will accept it. We are planning to determine the structure of a crystalline protein here, in the near future. I don't think that the job can be done by one man in a few years, and I am not sure that Dintzis is interested in collaborating on it.
We shall have to have one or more people who know the techniques of getting good x-ray photographs of crystals of proteins. We hope in a few months to have selected the crystals to be studied. I wish that I could talk with you in detail about our project - I do not want to write about it in detail, because I think that we should test out our ideas before talking about them. Please do not spread the word around. I have written Kendrew that we are planning to attack the problem.
Our rotating anode x-ray tube is standing idle. Sheldon Crane quit his job in 31 December. He has an industrial job that pays much more. I wish that we had somebody who would concentrate his attention on this x-ray tube, and perhaps other instrumental questions connected with protein photography. I think that something really exciting is going to happen here during the next year or two, and instrumentation could be of great value to us.
We had pretty well decided to look forward to having a spectrometer, in the course of time, for collecting three-dimensional protein crystal diffraction data. I agree with you that it would be nice if it were automatic. In the meantime, however, we are going to move forward with use of visually estimated intensities. You did not answer my question about the microphotometer giving integrated intensities to within 2%. I thought, from what Dintzis had said in his letter, that you had such a microphotometer in use in the Cavendish. Do you? How do you get the intensity values from your photographs of myoglobin?
Love from
