Met Dunitz while serving as Eastman Professor at Oxford University in 1948. Arranged for Dunitz to be given an appointment as Research Fellow at Caltech. Dunitz worked there for several years; he now works at the Public Health Service in Maryland. Dunitz is a first-rate crystallographer, characterized by thoroughness and originality. Dear Dr. Orlans:
I have been interested to read, in NATURE of 23 April 1955, your communication on the N-terminal amino acids of rabbit gamma globulin and antibody.
I am writing to ask about the penultimate sentence. You say that your results make it improbable that the part of the molecule containing the terminal groups is involved in either of these specific combining groups of the molecule. Am I right in my surmise that you feel that the specificity of combining groups is necessarily associated with a difference in amino-acid composition and sequence?
Fifteen years ago I proposed a theory of the structure and process of formation of antibodies in which the idea was presented that different specific combining groups represent different ways of folding of the polypeptide chain. I think that this theory remains worthy of consideration. I may mention that in this paper, a copy of which is enclosed, the suggestion was first made that precipitating and agglutinating antibodies are bivalent rather than multivalent. The bivalence has now received much experimental support.
