Dr. William B. Castle
Boston City Hospital
Boston
Massachusetts
Dear Bill:
I now have a graduate student (Harvey Itano, M.D.) beginning work on the problem of the relation between the nature of the hemoglobin in sickle cell anemia and the phenomenon of sickling.
He has not found any references in the literature to the work that you were telling me about, which, if I remember correctly, indicated that the dividing line between sickling and non-sickling came at 50 percent oxygenation of the hemoglobin or 50 percent combination with carbon monoxide. Could you tell me whether you and your collaborators have published any of this work, send me reprints if it has been published, and send me a brief statement about the results if it has not been published.
Last summer Dr. Burch told me that he felt sure that the phenomenon was due to a large amount of carbon dioxide. I have read his papers, and it seems to me that all of his results can be explained by assuming that the carbon dioxide treatment removes oxygen.
We are hoping to get some interesting results by studying other compounds of hemoglobin.
With best regards, I am
Sincerely yours.
Linus Pauling

Box 14-A, Station A
Ames, Iowa
Dear Dr. Spedding:
I thank you for your letter of November 7. I am very interested to learn that you have been having such good success with the rare earth program, and that you are planning to investigate rare earth metals and their alloys. Best wishes to you.
In view of this circumstance, we shall, I believe, give up the main program which we had planned, and carry out only a small amount of work on these metals, in order to test some special points.
In particular, we would like to make measurements of the lattice constant of cerium as a function of temperature, and also to reinvestigate the magnetic properties of the metal. It would be a great help to us to have some of the pure cerium which you have prepared, and I would be grateful if you could send us a supply, perhaps 25 grams.
I shall be very interested to learn the results that you obtain about the crystal structure of samarium, as well as all the other results that you and Dr. Rundle and coworkers obtain. I assume that your investigations on the properties and crystal structure of these metals and their alloys will be published without delay.
Sincerely yours,
Linus Pauling

[postmark on envelope reads: Nov. 23, 1946]
Dear love: I'm off in a few minutes to the Guggenheim meeting. Yesterday was a fine day. Warren Weaver looks very well - I saw him both before and after noon. He is unable to stay upright at night or with his eyes closed, and he weaves a bit as he walks. The Infantile Paralysis people also were very friendly. Warren said that he thinks that they want to do the whole job, and that he will encourage them; otherwise the R.Fn will help out. Also the two together will give us about $130,000 for 1946-7, while the big decisions are being made.
Luiy came at 6, and we had a Pizza on 48th St., and then went to the Mirsky's. Reba gave us some tea, with cheesecake for Luiy. Alfred says that Cori is without any doubt the leading chemical physiologist or biochemist in the

country, or that Gasser [sp ?] is trying to get him. Perhaps I'll drop a note to him. Also we should perhaps get Erwin Brand.
A strange thing happened yesterday. Henry showed me a set of wonderful photographs made be an applicant, Wayne Miller, of people along the street at Roosevelt's funeral, and I began to cry! Perhaps I'm not quite over the physical experiences of the last month. Of course, the emotional effect of that funeral was very great.
Henry is going to take us on a ride to his farm in Connecticutt on Sunday, April 28.
Love from
Paddy

Dear Linus:
How nice it is to have a word from you and probably to learn something more as a result of your work about that most interesting condition, sickle cell anemia. With regard to the facts about sickling, it is well established that oxygen and carbon monoxide prevent sickling, and that exposure to other gases produce sickling by the removal of one or the other of these. Naturally, there is some effect from carbon dioxide in so far as it alters the saturation curve for oxygen of the hemoglobin by a change in the pH of the system. I would agree with you that Doctor Burch's papers can all be interpreted in terms of removal of oxygen and, indeed, I wrote a critique of one of them for the Year Book of Medicine in which I simply interpolated the abstract of his communication in terms of this explanation.
I think that the literature that would be most useful for you, both for its content and for the references given, is the following:
Scriver, J. B., and Waugh, T. R. Canad. Med. Assn. J. 1930, 23, 375-380.
Murphy, R. C., and Shapiro, S. Arch. Int. Med., 1944, 74, 28. (Cf. Bibliography)
Same authors, Annals Int. Med. 1945, 23, 376. (Cf. Bibliography)
Our own observations here confirm those of Scriver and Waugh that sickling begins at about 35 to 40 millimeters oxygen tension. We were interested to determine the sickling by the effect on the "viscosity"; that is, the timed flow of blood through a viscosimeter with appropriate arrangements to maintain the blood in equilibrium with various tensions of oxygen. The viscosity of the blood begins to increase at about 40 millimeters oxygen tension, and rises
Dr. Linus Pauling, November 25, 1946—2
sharply in the range between 20 and 40. As you know, the tension of the capillary blood is in this range, and consequently the sickling phenomenon probably is the basis of the rather uniform and striking pathology of sickle cell disease. We have published only a sentence or two on this topic, but, as far as I know, it is the first observation of the "viscosity" of the blood. The reference is:
Ham, T. H. and Castle, W. B. Tr. Assn. Am. Phys., 1940, 55, 127.
I hope this is the information you want. It would be pleasant to see you sometime, and in the meantime I send all best regards to you.
Sincerely yours,
William B, Castle, M.D.
