Dear Dr. Pence:
I am very pleased that you are planning to go ahead at once with the preparation of the compounds that we need for our work in immunology. The method that I suggest for your obtaining the chemicals that you need is given in the postscript to this letter.
For a general survey of the field of immunochemistry I recommend Marrack's monograph, the title of which is given in a footnote at the beginning of my 1940 paper. I think it might well be available in the library of the University of Oregon Medical School, but you might want to order one for yourself from the British Library of Information, 270 Madison Avenue, New York City. It is not expensive—three shillings in England, and about a dollar, I think, here. Landsteiner's book is also worth having, but it is somewhat harder to understand than Marrack's. The usage of the word hapten is not uniform. Landsteiner introduced the word to refer to a molecule such as a carbohydrate which seemed to be important to the antigenicity of some coupled protein, but which by itself would not produce antibodies when injected into an animal. I use the word to refer either to the molecule or to the group when it is attached to a larger molecule. Sometimes I refer to the group as a haptenic group.
I think that your experiment of taking an antibody and turning it into another antibody in the test tube would be worth carrying out. The job of obtaining purified antibody is somewhat troublesome, so that we shall probably not try this experiment for some time.

Dear Professor Owens:
In answer to your question in your letter of May 29, I would say that I think it improbable that the proteins which combine with two or more different prosthetic groups to form different active enzymes do so as a result of modification of structure by the prosthetic group. I think that the same protein might well have such a structure that it has the power of combining with more than one prosthetic group. On the other hand, I think that it might well be possible under suitable circumstances for an enzyme to change its structure in such a way as to acquire new properties. We are planning some experiments now along these lines.

I am sorry that my answer to your letter of November 29, 1940 has been so long delayed. Because of a period of illness, I put a number of letters to one side, and have only now got around to answering them.
The work described in your letter and the accompanying reprint is very interesting. I feel, as do you, that your results are not incompatible with my theory of antibody structure. The variation in amount of destruction of antibody potency for different antisera probably is due, as you suggest, to differences in the stability of the antibody configuration.
You may have noticed the report of the work which Dr. Campbell and I have been doing on the manufacture of antibodies in vitro. Dr. Campbell has now tested a preparation of antiserum against Type I pneumococcus polysaccharide, and has found that it protects mice against Type I pneumonia.

I can now report progress on the carbon monoxide instrument. The lines of attack which I have though of have been followed up, but have not so far come to anything. However, about six weeks ago I told Dr. Arnold O. Beckman, formerly Assistant Professor of Chemistry in our Department, and now Head of the National Technical Laboratories, about the need for this instrument, and about ten days ago he came ot me with an idea which he thought would work. He has now made further calculations, with results that look very promising.
The instrument which he has in mind would work in the following way. Air would pass through a tube such that the absorption of a beam of light through the air could be measured, then through a layer of heated iodine pentoxide, which would be reduced by the carbon monoxide with formation of iodine. The air would then continue through another tube in which the absorption of light could be measured. The change in light absorption in a region where iodine vapor is absorbing, as measured by a split-element photovoltaic cell, would give the amount of carbon monoxide initially present in the air.
Using the 1936 data of Rabinowitch and Wood, for light with wavelength about 5000 Å, Dr. Beckman has calculated that it should be practicable to detect the presence of carbon monoxide in quantities as small as ten parts per million, that is, one thousandth of one per cent of an atmosphere. Larger quantities could presumably be determined with considerable accuracy. This calculation is based on the use of a cell one meter long, which presumably could be a tube bent around into a circle, with the light transmitted through it by internal reflection.
The volume of the apparatus as planned would be perhaps one-half cubic foot, its weight not over ten points, and the power source any convenient supply, 6 or 12 or 110 volts. The instrument would indicate amount of carbon monoxide continuously, and could be arranged to provide an alarm. The estimated price of the instruments in quantity production is a few hundred dollars.
I would be glad to know what the present situation is about a carbon monoxide instrument. If there is still the need for the instrument described in your letter, I would recommend that Dr. Beckman be given a contract for developing an instrument along the line which he has suggested. From the success which he has had with his pH meter and spectrophotometer, I would expect him to do a good job.
